Microglial stress response in Alzheimer’s disease progression plays a pivotal role in driving neurodegeneration. Activation of the integrated stress response (ISR) in microglia leads to toxic lipid secretion and synapse loss. This cellular stress pathway exacerbates pathology in disease models, while pharmacological inhibition of ISR or lipid synthesis shows protective effects.
